Zusammenfassung
1The effect of the chaotropic agents on three different preparations of the sarcoplasmic reticulum, i.e. native membranes, membranes whose phospholipids were hydrolyzed by phospholipase A and lipid-deficient membranes has been studied. 2The chaotropic anions inhibit calcium transport, calcium-dependent ATPase activity, phosphoprotein formation and ATP binding.The effectiveness of the anions increases in the following order: image 3Inhibition occurs immediately and is reversible up to an incubation period of 7 min. 4The maximal effective concentration of the chaotropic anions (0.4 M) does not solubilize either membrane protein or membrane lipids. 5The chaotropic anions increase the calcium-permeability of the sarcoplasmic membranes a relatively small amount, so that the permeability increase cannot account for the reduced calcium uptake. 6The interference between the binding of ATP and chaotropic anions is the most likely mechanism of their action.
