 Zusammenfassung: The background of a new project Monitoring and Control of High Density Cell Culture Systems is presented. The aim is the development of integrated concepts to design and optimize vaccine production processes. Besides the amelioration of viral yields and reproducibility of these processes our focus is the establishment of downstream processing methods to improve efficacy, purity and safety of vaccines. With a process of equine influenza virus production as an example we investigate: (a) Cultivation and scale-up of animal cells (MDCK) using microcarrier systems (b) Replication of influenza viruses (equine influenza Newmarket 1/93 H3N8) (c) Downstream processing to subunit or split vaccines. Within this concept the aim is to characterize options, advantages and limitations of high density animal cell culture technologies. Based on mathematical models describing cell growth and virus replication in bioreactors, different control strategies will be established and the results will be compared with existing cultivation techniques.
