 Zusammenfassung: Understanding and predicting the performance of a chromatographic separation process primarily requires reliable information on the distribution of the components of a mixture between the mobile and stationary phases under equilibrium conditions. In this work we discuss results of studying the adsorption of Lysozyme on modified Sepharose matrices (STREAMLINE SP, Amersham Biosciences). Experiments were carried out with (a) packed beds and (b) in the expanded bed mode. Frontal analysis has been used to determine the adsorption isotherms from different experimental runs with the packed bed. The isotherms were fitted using both a Langmuir as well as a rectangular isotherm. The isotherms were verified by predicting further fixed bed breakthrough curves and comparing the results with experiments. Subsequently, the porosity and the dispersion coefficient of an expanded bed were estimated analyzing selected experimental results for one feed concentrations. Finally, all parameters determined were used to predict the performance of the expanded bed for different feed concentrations.
