 Abstract: Abstract Two methods were tested for their suitability in quantitative investigations of sediment bacteria in sandy freshwater deposits. After homogenizing the sediment (homogenizer type "Ultra-Turrax TP 18/2 N") it was possible to determine the number and biomass of bacteria by epifluorescence microscopy. It was impossible to determine the abundance of bacteria by scanning electron microscopy. There were only limited possibilities to investigate the distribution of attached bacterial cells on sand particles. On Nov. 30, 1979 2.0 * 109 bacteria per ml sediment and 0.045 mg bacterial biomass (dry weight) per ml were found in the surface sediment of the Breitenbach, a small grassland stream in eastern Hesse. The bacterial biomass corresponded to 1.1 % of the total organic matter in the surface sediment.
